INCI: Tetrapeptide-21
Category: Signal Peptide / Collagen-Stimulating Active
Used in: Serums, premium anti-ageing moisturisers, eye treatments
Typical Usage Level (Topical): 0.5-5% as part of a commercial blend.
What This Ingredient Does
Tetrapeptide-21 is a newer generation signal peptide that stimulates the production of collagen I, collagen IV, and fibronectin key structural proteins in both the dermis and the dermal-epidermal junction. Its mechanism is similar to Matrixyl-type peptides in that it communicates with fibroblasts to upregulate matrix protein synthesis, but independent studies have shown it to outperform Matrixyl 3000 in collagen-stimulating activity at comparable concentrations. It also supports the basement membrane zone, which is responsible for skin plumpness and bounce at a deeper structural level than surface collagen alone.
Key Benefits
-
Stimulates collagen I, IV, and fibronectin, addressing both surface lines and deeper structural loss
-
Outperforms Matrixyl 3000 in collagen production in comparative in vitro studies
-
Supports the dermal-epidermal junction for improved skin plumpness and bounce
-
Improves wrinkle volume and skin density with consistent use
-
Well-tolerated; no known irritation or sensitisation
Who It's Best For
-
Those 35+ with moderate skin ageing volume loss, deeper lines, reduced density
-
Users already familiar with Matrixyl peptides looking for an upgrade
-
Suitable for all skin types including sensitive skin
-
Works well combined with Retinol or HPR for multi-pathway collagen support
Clinical Note by Dr. Su
Tetrapeptide-21 has not yet reached the consumer awareness that Matrixyl has, but the data behind it is credible. If you are already using a Matrixyl 3000 product and want to step up, this is a logical next ingredient to look for. Give it at least 10–12 weeks before assessing results.
